ARTIFICIAL AND NATURAL NUCLEIC ACIDSELF ASSEMBLING SYSTEMS byMARCUS WOODAugust 2011Advisor: Dr. John SantaLucia, Jr., Dr. David RuedaMajor: ChemistryDegree: Doctor of Philosophy Nucleic acids are good candidates for nanomachine construction. They participatein all the processes of life, and so can function as structural building blocks and dynamiccatalysts. However, to use nucleic acids as nanomachines, a better understanding of theirmaterial properties, how to design structures using them, and their dynamics is needed. We have tried to address these issues, in a small way, with nucleic acid force fielddevelopment, an attempt at nanostructural design and synthesis using DNA, and a studyof the RNA/protein regulatory dynamics of the tryptophan regulatory attenuation protein.

Sterically stabilized self-assembling reversibly cross-linked polyelectrolyte complexes with nucleic acids for environmental and medical applications.

One of the principal problems facing nucleic acid delivery systems using polyplexes is the instability of the complexes in the presence of proteins and high salt concentrations. We have used a cross-linking polymer to overcome this problem. Pendant thiol moieties have been incorporated into a PAA (polyamidoamine) homopolymer and a PEG [poly(ethylene glycol)]-PAA-PEG copolymer reported previousl...

Using DNA to program the self-assembly of colloidal nanoparticles and microparticles

Deoxyribonucleic acid (DNA) is not just the stuff of our genetic code; it is also a means to design self-assembling materials. Grafting DNA onto nanoand microparticles can, in principle, “program” them with information telling them exactly how to self-assemble. Although fully programmable assembly has not yet been realized, the groundwork has been laid: with an understanding of how specific int...

Design of interacting multi-stable nucleic acids for molecular information processing

Despite an exponential increase in computing power over the past decades, present information technology falls far short of expectations in areas such as cognitive systems and micro robotics. Organisms demonstrate that it is possible to implement information processing in a radically different way from what we have available in present technology, and that there are clear advantages from the pe...

The effect of polylactic acid support in stability and electrical field of heterocyclic coupled hexa peptide nano systems: A novel strateu to drug delivery

Biological materials. recently. are the building blocks of several self-assembling peptide and protein systems. The main challenge In molecular self-assembly is to design molecular building blocks that can undergospontaneous organization. These cyche peptides were produced by an alternating fl'ell number of D- and Laminoacids.which interact through non-covalent interactions co an array of selfas...

Adsorption of Polylysine, Poly(glutamic) acid and their Block Copolymers on Polystyrene and on Carbon Nanotubes

A better understanding of protein behavior at solid-liquid interfaces is important to improve our knowledge of various natural and synthetic processes. A few examples where protein adsorption at a solid-liquid interface may play an important role are found in: blood coagulation, artificial kidney failure, biomedical microdevices, self-assembling microelectromechanical systems (MEMS), protein ba...
